• The zero-carbon background enabled isolation and collection of hyphae-secreted GRSP. • Purified GRSP contained peptides homologous to Hsp60 and exhibits a network-like microstructure. • DE-GRSP showed higher hydrophobicity and Fe2+ valence than EE-GRSP; both have humic-like spectra. • Fungal Hsp60s were differentially upregulated in symbiosis, and FmHsp60 was induced more strongly. • GRSP was a functional molecule with defined structure, not an inert microbial residue.
