• METTL3-dependent m6A modification promotes macrophage efferocytosis under inflammatory stress. • Mettl3 deficiency impairs apoptotic cell clearance and aggravates chronic inflammatory injury. • METTL3 stabilizes MFGE8 mRNA through IGF2BP2/3-mediated post-transcriptional regulation. • Defective efferocytosis contributes to inflammation-associated colorectal carcinogenesis. • MFGE8 supplementation restores efferocytosis and reduces tumor burden in the AOM/DSS model.
