• Estrogen deficiency depletes gut microbiota-derived deoxycholic acid (DCA) in PMOP. • Mendelian Randomization identifies DCA as a causal protector of bone density. • DCA supplementation effectively mitigates OVX-induced trabecular bone loss in vivo. • DCA restores bone homeostasis by promoting BMSC osteoblastogenesis over adipogenesis. • DCA inhibits osteoclastogenesis by suppressing the IκB-α/NF-κB/NFATc1 signaling pathway.
