• Cardiovascular disease continues to be the leading cause of death worldwide. • Diosmin improved vascular function by reducing VCAM-1, ICAM-1, as well as p-JNK and p-NF-κB proteins. • Diosmin and diosmetin exerted antioxidant and anti-apoptotic effects by increasing the ratios of phospho-AKT to AKT and phospho-Nrf2 to Nrf2. • Diosmetin has the capacity to reduce autophagy through the inhibition of the PI3K/Akt signaling pathway and endothelial-mesenchymal transformation via the Src-targeting pathway. • The collected findings emphasize the role of diosmin and diosmetin in safeguarding against heart failure, atherosclerosis, hyperlipidemia, and vascular dysfunction. • This addition provides a novel therapeutic and preventive choice for cardiovascular disease.
