Discovery of rare cycloartane-type triterpenoid sulfates from Wrightia pubescens and their neuraminidase inhibition activity

Ten undescribed cycloartane-type triterpenoids (1-10) including six rare sulfate complexes (2-3, 6, 8-10) were isolated from the roots of Wrightia pubescens. Their structures were unequivocally determined through comprehensive analysis of NMR spectroscopic data and single-crystal X-ray diffractions (1-3, 6-7). Additionally, transmission electron microscopy combined energy-dispersive X-ray spectroscopy verified the presence of sulfur and calcium in sulfate complexes (2-3, 6, 8-10). Sulfated triterpenoids 2-3, 6, 8-10 showed significant neuraminidase (NA) inhibitory effects with IC50 values ranging from 8.44 mu M to 73.46 mu M. The dissociation constants of compounds 2-3, 6 binding to the NA protein were measured by surface plasmon resonance as 1.7, 4.3, and 6.0 mu M, respectively. These binding affinities were further confirmed by molecular docking analysis. These results suggested compounds 2-3, 6 have potential utility for the treatment of influenza virus infection.
