• HDAC2 epigenetically silences DLX2 by reducing H3K9ac at its promoter. • DLX2 knockdown attenuates VSMC calcification and glycolysis in vitro and in vivo. • The pro-calcific effect of DLX2 is mediated via WNT1/β-catenin-glycolysis axis.
