• We tested sex/pubertal effects on brain response to emotional faces in preadolescence. • Sex differences in brain response to emotional faces were not found. • Any one puberty marker alone was unrelated to brain response to emotional faces. • The effect of estradiol on ACC response to happy faces varied by pubertal stage. • Studying the confluence of pubertal stage and hormones on brain development is needed.
