• Nobiletin alleviates slow-transit constipation by restoring gut barrier integrity and mucus secretion. • It modulates bile acid signaling via the FXR/TGR5-ASBT in ileum and FXR/TGR5-5-HT3R in colon. • Nobiletin remodels gut microbiota, reversing dysbiosis. • Treatment rebalances plasma neurotransmitters and enhances intestinal propulsion in constipated mice.
