Effects of curcumin on inflammatory and oxidative stress related markers in rat models of experimental arthritis: a systematic review and meta-analysis

This study evaluated whether orally administered pure curcumin affects inflammatory cytokines and oxidative stress-related markers in rat models of experimental arthritis. We searched Web of Science, PubMed, Embase and Scopus for studies published up to June 2026. Studies used rat models of experimental arthritis, compared orally administered pure curcumin with vehicle or untreated controls, and reported inflammatory or oxidative stress-related outcomes. Model selection was based on heterogeneity: fixed-effect analyses were performed for outcomes with I² values of 50% or lower, while random-effects analyses were conducted when I² exceeded 50%. Ten studies were eligible for inclusion. Compared with control conditions, curcumin administration was associated with decreases in IL-1β (SMD = − 2.91, 95% CI: −3.59 to − 2.22) and TNF-α (SMD = − 1.50, 95% CI: −1.97 to − 1.04), while IL-10 levels were increased (SMD = 1.23, 95% CI: 0.34 to 2.13). Curcumin also improved oxidative stress-related profiles, with higher levels of SOD (SMD = 1.13, 95% CI: 0.12 to 2.15), GSH (SMD = 1.48, 95% CI: 0.65 to 2.30), CAT (SMD = 1.57, 95% CI: 0.47 to 2.66), and GPx (SMD = 2.94, 95% CI: 0.54 to 5.35), and lower LPO levels (SMD = − 0.94, 95% CI: −1.56 to − 0.32). Sensitivity analyses supported the direction of the findings. Orally administered pure curcumin was associated with favorable changes in inflammatory and oxidative stress-related markers in rat models of experimental arthritis. These findings warrant further well-designed animal studies using standardized methods and clinically relevant structural, functional, and safety outcomes.
