Effects of dietary organic trace elements on the health status in shell gland of laying hens replacing of inorganic sulphate source

The experiment aimed to investigate the effects of organic trace elements (OTE) on shell gland health in laying hens challenged with salpingitis. A total of 40 Jingfen - 1 hens (33 weeks old) were randomly assigned to 4 groups: inorganic trace elements (ITE), OTE, ITE + salpingitis model (ITE_SM), OTE + salpingitis model (OTE_SM). Diets contained 60 mg Zn/kg and 80 mg Mn/kg from either sulfate salts (ITE) or yeast zinc and manganese methionine (OTE). After 9 weeks, salpingitis model was conducted in SM groups. Results showed that under SM treatment, the significant changes in the inflammatory response enhancement, immune function suppression, and related indicators of shell gland morphology and function indicate the successful construction of the SM in laying hens (P < 0.05). Compared with ITE, OTE supplementation enhanced Zn content, improved mucosal morphology, upregulated barrier-related genes in shell gland, elevated serum IgA content, reduced pro-inflammatory cytokines level in serum, suppressed inflammatory gene expression and eggshell formation-related genes in the shell gland (P < 0.05). The carbonic anhydrase activity in serum and the zinc content and the mRNA expressions of Claudin2, p38 mitogen activited protein kinases, Tumour necrosis factor-α, carbonic anhydrase, and Ovocledidin-116 in shell gland, significant interactions were observed between the SM and TE sources (P < 0.05). In conclusion, these findings indicate that replacing inorganic sulfates with organic Zn and Mn improves shell gland health via enhanced mineral retention, barrier integrity and matrix protein synthesis by suppressing the inflammatory response.
