• Orally administered EP395 reduces local neutrophil counts and cytokines in murine acute respiratory inflammation models. • Increased concentrations of the epithelial collectin, SP-D in LPS-stimulated lung washings were also reduced by EP395. • EP395, like azithromycin, demonstrates both acute stimulatory and inhibitory effects on neutrophil function in vitro. • EP395 represents a novel, non-antibacterial anti-inflammatory macrolide for COPD therapy.
