• 20-ring polyamides with guanidinium and tetramethyl guanidine groups have been synthesized. • Head group substitutions significantly weaken DNA binding for the 20-ring hairpin polyamide. • Head group substitutions perturb microscopic binding kinetics but not antiviral activity.
