• gadC was cloned into pETDuet-1-gadBD304G/F433L to construct co-expressing strain. • Whole-cell biocatalysis was optimized by single-factor and orthogonal experiments. • 382.59 g/L GABA was produced by a whole-cell catalyst recycling for 5 batches. • This study contributes to the industrial production of GABA by engineered strain.
