• Damaged gut barrier function, ovarian inflammation, and estradiol secretion disorder are observed in PCOS mice. • GLP-1 notably enhances the gut barrier function and alleviates ovarian inflammation. • ETA improves PCOS by regulating GLP-1 levels to restore the intestine and block the ovarian inflammation condition. • Reveal the mechanism by which GLP-1 affects PCOS dual-action for the first time.
