• Nano delivery systems significantly enhance the efficacy and safety of CAR-T and CRISPR-Cas9–based cancer therapeutics. • Nanocarriers enable targeted, non-viral delivery of CAR and CRISPR components, minimizing off-target effects and immunogenicity. • Integration of nanotechnology improves in vivo stability, transfection efficiency, and controlled release of gene-editing payloads. • Combined Nano-CAR-T and Nano-CRISPR approaches hold promise for overcoming tumor resistance, antigen escape, and toxic manifestations such as cytokine release syndromes. • Translational advancements in nanomedicine pave the way for next-generation, personalized cancer immunotherapy platforms.
