• Hierarchical miRNA-CaCO₃@PLGA nanoparticles are nanoassembled with miR-124-loaded CaCO₃ cores encapsulated by PLGA. • Lysosomal pH-triggered CaCO₃ decomposition co-releases Ca²⁺ and miR-124 for synergistic pro-neurogenic effects. • In vitro, hierarchical miRNA-CaCO₃@PLGA nanoparticles enhance NSC neuronal differentiation to 32.4% vs 16.7% spontaneously. • Transplanted NSCs endocytosing miRNA-CaCO₃@PLGA nanoparticles promote cognitive and motor recovery in TBI mice.
