• The synthesis of a novel biocompatible MOF based on 2-aminopurine. • The synthesis of a pH-responsive prodrug with CHO modification of MTX and covalently attachment to the NH₂ groups of the bio-MOF. • The in vitro cumulative release evaluation in PBS solutions at pH of 5.0 and 7.4 to assess the role of the pH-sensitive CN bond.
