Engineering strategies should be tailored to the specific properties of both the cell type and the nonliving drug. Engineering cells with nonliving drugs confers resistance to microenvironmental reprogramming and enhances tumor penetration. Controlled drug release helps preserve cellular function. Microenvironment-responsive triggers enable the convenient release of surface-bound cargo, whereas exocytosis-controlled modules are well suited for intracellular cargo loading. Safety risks and product heterogeneity remain key challenges for clinical translation.
