• In situ TEM micro-tensile experiments reveal orientation-dependent plastic deformation pathways in single-crystalline CrCoNi medium-entropy alloys. • Twin boundary migration (TBM) and twin boundary sliding (TBS) are directly distinguished and shown to operate sequentially or cooperatively depending on local deformation conditions. • A cooperative TBM–TBS deformation mode is identified and enabled by local stress concentration effects beyond crystallographic orientation constraints. • Twin–twin interactions are revealed to play a dual role in activating and stabilizing TBM–TBS coupling, enabling sustained plastic strain accommodation.
