• Hepatic ENT1 expression exhibits stage-specific differences during MASLD progression in mice and humans. • ENT1 deficiency induces mild basal metabolic alterations in the liver. • ENT1 loss protects against diet-induced liver disease in association with reduced body weight and improved metabolic status. • Hepatic ENT1 expression correlates with insulin levels and HOMA-IR in humans with early MASLD. • Nucleoside transport emerges as a regulatory axis in metabolic liver disease.
