• Elaeocarpus sylvestris exhibits anti-inflammatory activity in LPS-stimulated keratinocytes. • Geraniin was identified as a major constituent of ESLE by HPLC analysis. • ESLE modulates MAPK, NF-κB, and JAK/STAT signaling, indicating its role in inflammatory responses. • ESLE alleviates AD-like symptoms, suggesting improved skin barrier function in vivo. • This study provides pharmacological evidence supporting the traditional use of E. sylvestris in skin inflammation.
