• Dynamic Fgf8 and receptor expression profiles define a specific regulatory window in early mandibular development. • FGF8 promotes mandibular cell expansion at E10 while simultaneously suppressing chondrogenic differentiation. • The pro-proliferative response to FGF8 is restricted to E10 and abolished at E11 due to an autonomous transcriptional shift. • FGF8-induced inhibition of Meckel’s cartilage formation is mediated by and dependent on the MEK/ERK signaling pathway. • FGF8 acts as a stage-specific regulator that coordinates the balance between cell proliferation and chondrogenic commitment.
