Evolution-informed pest management (EIPM) leverages evolutionary principles to anticipate pest and pathogen adaptation, enabling durable crop protection. Evolutionary analysis identified a PUB21DN allele in resistant citrus relatives. PUB21DN functions as a loss-of-function allele that confers citrus resistance to Huanglongbing (HLB) by stabilizing MYC2 and activating plant immunity. In susceptible cultivars, a helitron element inserted into the PUB21 promoter enhances its expression and accelerates MYC2 degradation, thereby increasing HLB susceptibility. Artificial intelligence (AI) designed the micropeptide APP3-14, which works by inhibiting PUB21 to stabilize MYC2 and boost immunity, while also directly killing the Candidatus Liberibacter asiaticus (CLas). Integrating AI, synthetic biology, and gene editing offers a blueprint for developing biopesticides and HLB-resistant citrus cultivars.
