• FKBP10 is downregulated in osteoporosis, and FKBP10 overexpression attenuates OVX-induced bone loss. • FKBP10 enhances osteogenic differentiation of BMSCs by increasing ALP activity and mineralization. • FKBP10 directly interacts with HSPA5 and promotes HSPA5 lysosomal degradation. • FKBP10 alleviates ERS-mediated pyroptosis to preserve the osteogenic capacity of BMSCs.
