Gas chromatography - tandem mass spectrometric determination of bisphenols from herbal food formulations by Ion-pair solid-phase extraction and in-situ methylation method

Bisphenols are among the significant classes of endocrine-disrupting chemicals (EDC) that mimic the human endocrine system, causing hormonal dysfunctions. In the present work, an ion-pair solid-phase extraction (SPE) method followed by in situ methylation and gas chromatography-tandem mass spectrometric (GC-MS/MS) determination of 13 bisphenols for trace-level detection in herbal food formulations was developed and validated. The analytes were converted to methyl ethers in the GC injection port, yielding good peak shapes and enhanced sensitivity compared to direct analysis. The method showed linearity over the range of 1 to 5000 ng/g, with an R-2 value greater than 0.99. The inter-day and intra-day precisions measured at three concentration levels, 25, 250, and 2500 ng/g levels, were below 15%. The recovery of the analytes from the herbal food products ranged from 95% to 105%, with matrix effects <10% at these concentration levels.
