• Lattice relaxability, modulated by microstructural strain and defects, governs self-trapped exciton formation and thermochromism in 2D (PEA)2PbBr4. • Cooperative lattice relaxation, essential for exciton self-trapping, is suppressed in polycrystalline thin films by synergistic strain-defect pinning. • Engineering the strain-defect landscape offers a strategy to gate excitonic pathways and enable stimuli-responsive functionalities in layered perovskites.
