• 25 PRN genes were genome-wide identified and characterized in Arabidopsis and seven Rosaceae. • PbrPRN2 and PbrPRN3 were identified as pollen-associated candidate genes in pear. • Altered expression of PbrPRN2 and PbrPRN3 affected pear pollen tube elongation. • PbrPRN2 and PbrPRN3 may regulate pollen tube growth through flavonol–ROS homeostasis.
