• Sediments are the source of 2-MIB release in Xuhu (Lake Taihu), with filamentous cyanobacteria (Planktothricoides, Pseudanabaena; Dolichospermum minor) confirmed as primary producers via gene markers and 2-MIB cyclase activity. • Sedimentary GPP is the key precursor: regions with GPP-rich sediments exhibit significant 2-MIB release (up to ∼17 ng L⁻¹), whereas GPP-poor regions show negligible release. • Species-dependent responses: exogenous GPP boosts 2-MIB per biomass most strongly in Dolichospermum, moderately in Pseudanabaena, and least in Planktothricoides. • Strong GPP-macrophyte linkage: sediment GPP levels are strongly correlated with the distribution and decay of submerged macrophytes across lake regions. • Operational implication in macrophyte-rich shallows: targeted removal of macrophyte residues (especially after winter die-off and wind-induced resuspension) can reduce sediment GPP pools and lower the probability of T&O episodes.
