• Glycan under-sulfation boosts model antigen presentation by lymphatic endothelium. • CD8+ T cell activation is augmented by antigen-pulsed glycan targeted lymphatics. • This state drives increased lymph node CD8+ T cells with reduced metastases in vivo. • Glycan targeting of lymphatic endothelium may augment anti-cancer adaptive immunity.
