• MTX and DEX modulate NLRP3-related gene expression and HLA class I expression in primary human retinal pigment epithelial cells under basal and H₂O₂-induced oxidative stress conditions. • Changes in transcriptional profiles of NLRP3- and antigen-presentation–related genes under H₂O₂-induced conditions are not paralleled by proportional alterations in IL-1β secretion. • Combination therapy is associated with reduced IL-18 secretion under oxidative stress conditions.
