• Results systematically characterize myocardial injury and cardiac electrophysiological abnormalities caused by heat stroke. • Heat stress mediates mitochondrial dysfunction through the Ca2+/Calcineurin/p-DRP1(Ser637) signaling axis leading to myocardial injury and ventricular arrhythmias. • MitoTEMPO and FK506 ameliorates heat stroke-induced myocardial injury and ventricular arrhythmias.
