• Ring-expanded α,β-unsaturated ketone is designed as selective Cys response site. • Py-xan exhibits superb fluorescence enhancement (> 300 folds) for Cys detection. • Broad linear range (0.5-250 μM) detection of Cys is achieved by the Py-xan. • This fluorescent probe is applied to Cys imaging in multiple living systems.
