• Hepatic HSF1 levels are regulated by ATF3 and decreased during MASLD. • HSF1-LKO mice aggravates, while HSF1-LOE protects mice from MASLD via Pparα. • Hsf1 activation regulates Uchl1 transcription to enhance Pparα protein stability. • Uchl1-LOE and fenofibrate rescued Hsf1 deficiency caused hepatic lipid dysfunction.
