• HYDRA-EDGE is a portable POCT biosensor fusing DNA hydrogels, microfluidics, and glucose sensing. • The engineered bi-legged DNA walker drives enzyme-free cascade amplification, delivering ultrahigh sensitivity beyond single-leg systems and commercial kits. • The DNA hydrogel matrix ensures spatial confinement, stable enzyme encapsulation, and ambient stability for field use. • Clinically validated against RT-PCR, it accurately detects influenza A/B and SARS-CoV-2 with simple handling and low cost, enabling robust on-site multiplex diagnostics.
