Sulfotransferases (SOTs) catalyze sulfate conjugation and contribute to diverse biochemical processes in plants. In this study, 112 wheat SOT genes were identified and characterized through genome-wide analyses. The TaSOT genes were unevenly distributed across 21 chromosomes, and duplication analysis revealed 35 duplicated gene pairs that have undergone purifying selection. Phylogenetic and synteny analyses showed that TaSOT proteins are more closely related to SOTs from barley and rice than to those from Arabidopsis. Expression analysis revealed that TaSOT genes respond to abscisic acid (ABA), hydrogen peroxide (H2O2), indole-3-acetic acid (IAA), Fusarium graminearum infection, and zebularine treatment. TaSOT48 was selected for functional validation based on its consistent induction across treatments. Heterologous expression of TaSOT48 in yeast enhanced growth under ABA, IAA, H2O2, drought, and salt treatment conditions, whereas silencing TaSOT48 in wheat increased susceptibility to F. graminearum, accompanied by elevated MDA levels and reduced POD, CAT, and SOD activities. Transient expression confirmed that TaSOT48 localizes to the cytoplasm. These findings provide a framework for understanding wheat SOT genes and identify TaSOT48 as a promising candidate for improving stress resilience in wheat.
