• ICIs are reshaping SoC for resectable LA-HNSCC. • Phase III KEYNOTE-689 trial showed perioperative pembrolizumab adds benefit to SoC. • Phase III NIVOPOSTOP trial with nivolumab plus SoC met its primary endpoint. • Further study is needed to optimize immunotherapy in the adjuvant setting. • Further study is needed for immunotherapy in unresected LA-HNSCC.
