• The prediction of B-cell epitopes is essential for advancing antibody therapeutics. • Computational prediction methods enable the rapid screening of large datasets. • Progress in machine learning substantially improved epitope prediction. • In spite of the recent progress, the current methods still require experimental validation. • The key to higher accuracy is improving the structural modeling of antibody–antigen interactions.
