• Heat stress (42℃) rapidly triggered leaf wilting/rolling, photosynthetic inhibition, membrane damage, and ROS accumulation in Lycoris radiata (L. radiata). • RNA-seq identified LrDREB2B (DREB-A2) as a heat-inducible regulator. • LrDREB2B localized to the nucleus and showed transcriptional activation activity. • LrDREB2B positively contributes to thermotolerance. • LrDREB2B modulated transcripts of the HSF-HSP pathway genes, antioxidant/ROS-scavenging genes, and GolS1, linking it to proteostasis, redox buffering, and galactinol-associated protection.
