• 1.First integration of 680 nm NIR-driven oxygen-tolerant MB/Cu photo-ATRP with Y-shaped DNA/CHA for electrochemical miRNA-21 sensing. • 2.Achieves ultrasensitive LOD of 53 aM and wide linear range (0.1 fM–10⁵ fM) in open air, with ~99% dissolved oxygen elimination within 40 min without deoxygenation. • 3.Dual amplification yields excellent specificity, reproducibility (RSD < 3.7%), and 10% serum compatibility. • 4.Provides simple, biocompatible, and scalable platform for preliminary POCT of tumor biomarkers in diluted samples.
