• TRM45540 controls pepper anthracnosein vitro and fruit trials. • OSMAC strategy confirms 5294 medium as optimal for TRM45540 to inhibiting Gloeosporium piperatum. • Pimaricin strongly inhibits G. piperatum. • Soluble starch-glucose-glycerol and yeast extract-tryptone boost TRM45540 biomass and activity. • It is stable at pH 7–9/1–2 h UV/∼20 °C can control effect comparable to chemicals.
