T cell dysfunction restrains the efficacy of immune checkpoint blockade and CAR-T cell-based cancer therapies. Iron-mediated ferroptosis is now shown to contribute to the dysfunction of endogenous T cells and CAR-T cells, suggesting that inhibition of ferroptosis could improve the anti-tumor activity of CAR-T cells in cancer.
