• ITGA11 promotes lens epithelial-mesenchymal transition via the PTGS2 axis. • ITGA11 depletion prevents metabolic reprogramming and inflammatory signaling. • Lipidomics reveals a PTGS2-mediated arachidonic acid-to-prostaglandin shift during lens EMT. • Pharmacological inhibition of PTGS2 effectively reverses the fibrotic phenotype.
