• CD6 expression pattern in MS enables selective targeting of pathogenic memory cells. • CD6 blockade mainly restrains inflammatory CD4+ T-cells diapedesis across the BBB. • CD6 blockade stabilizes BBB endothelial architecture and junctional protein levels. • CD6 blockade inhibits activation and tissue-residency programs in CD4+ T cells. • CD6 blockade potentially shields oligodendrocytes from direct T-cell injury.
