• Keratinocyte-derived exosomes mediate TGF-β1-induced activation of human dermal fibroblasts. • TGF-β1 promotes the packaging of LINC01605 into keratinocyte-secreted exosomes. • Exosomal LINC01605 enhances fibrotic phenotypes in fibroblasts by acting as a ceRNA for miR-370-3p to upregulate TGFBR2. • LINC01605 establishes an amplification mechanism to amplify and sustain the pro-fibrotic TGF-β1/Smad signaling pathway. • Blocking exosome secretion with GW4869 alleviates skin fibrosis in a mouse model.
