• LMBV infection enhanced aerobic glycolysis in vitro and in vivo. • Glucose and lactate were essential for efficient LMBV replication. • Lactate promoted LMBV replication by boosting cholesterol biosynthesis. • This study revealed a novel lactate-cholesterol-IFN axis in iridovirus pathogenesis.
