• First verified LIG directly binds STING with high affinity, and acts as a novel natural STING agonist. • LIG reverses NNK-induced cGAS-STING pathway suppression to exert lung cancer chemopreventive effects. • LIG triggers STING Golgi translocation and downstream signaling, uncovering natural product-mediated STING regulation.
