• Mass spectrometry shows no phosphorylation of phospholamban (PLB) in PLB-DM mice. • PLB-DM mice do not show remodeling to compensate for loss of PLB phosphorylation. • In-depth analysis of Ca2+ handling uncovers role of PLB phosphorylation in the heart. • Loss of PLB phosphorylation lowers SR Ca2+ load and release. • PLB-DM myocytes show blunted adrenergic response with slow Ca2+ reuptake kinetics.
