• Oil-based biphasic microfluidic sensing is implemented using commercial Tee assemblies and perfluorinated tubing. • An in-situ fluorescence detector is developed for real-time monitoring of fluorescence signals in segmented-flow microfluidics. • Channel deformation and dye adsorption are minimized compared to conventional PDMS microfluidic chips. • The microfluidic platform is applied to optical sensing of ions, albumin, and hydrogen peroxide.
