• Lrp10+/- mouse is animal model of LRP10 haploinsufficiency-induced PD. • Lrp10+/- mice display PD motor impairment and demise of SN dopaminergic neurons. • Lrp10 deficiency elevated α-synuclein mRNA and protein in SN of Lrp10+/- mice. • Lrp10 insufficiency activates SN ER stress and mitochondrial apoptotic cascades. • Lrp10 deficiency causes activation of microglia and microglial NLRP3 inflammasome.
