• Lycorine hydrochloride (LH) inhibits neointimal hyperplasia in vivo. • LH suppresses VSMC phenotypic switching via MAPK pathway inhibition. • Targeted binding of LH to Erk1 in the regulation of VSMC phenotypic switching. • LH shows potential as a pharmacological intervention for restenosis.
